Olaparib is a PARP inhibitor with the chemical name 4-[(3-{[4-(cyclopropylcarbonyl)piperazin-1-yl]carbonyl}-4-fluorophenyl)methyl]phthalazin-1(2H)-one. Its molecular formula is C24H23FN4O3 and its molecular mass is 434.46. Olaparib is a crystalline solid that is non-chiral and has low solubility regardless of pH. Lynparza tablets contain either 100 mg or 150 mg of olaparib as the active ingredient. The inactive ingredients in the tablet core include copovidone, mannitol, colloidal silicon dioxide, and sodium stearyl fumarate. The tablet coating is composed of hypromellose, polyethylene glycol 400, titanium dioxide, ferric oxide yellow, and ferrosoferric oxide (only in the 150 mg tablet).
Lynparza is a medication indicated for various types of Cancer. It is used for the maintenance treatment of adult patients with advanced ovarian, fallopian tube, or primary peritoneal cancer who have a BRCA mutation and are in complete or partial response to platinum-based chemotherapy. It can also be used in combination with bevacizumab for the maintenance treatment of advanced ovarian cancer in patients who have homologous recombination deficiency (HRD)-positive status. Additionally, Lynparza is indicated for the maintenance treatment of recurrent ovarian cancer with a BRCA mutation, the adjuvant treatment of high-risk early breast cancer with a BRCA mutation, and the treatment of metastatic breast cancer with a BRCA mutation. It is also used for the maintenance treatment of metastatic pancreatic adenocarcinoma and for the treatment of metastatic castration-resistant prostate cancer with an HRR gene mutation, either alone or in combination with abiraterone and prednisone or prednisolone. Patients should be selected for therapy based on an FDA-approved companion diagnostic for Lynparza.
